Police say four cars collided on the A4 Belfast Road at around 5.30pm.
Eight other people, including three children, were taken to hospital for treatment for their injuries that are not believed to be life-threatening.
John McPoland, from the ambulance service, said 11 emergency crews were dispatched to the scene.
On Friday night, he told UTV: "At the scene we found a number of patients who needed hospital treatment, and have taken them to the South West Acute Hospital and also Craigavon Area Hospital.
He added: "Because of the remote rural location, a number of local people in the area came out to the scene and attended to the casualties while the ambulance crews arrived and I would like to thank those people for their help."
Emergency services later stood down the major incident in the area.
Police have appealed to anyone who witnessed the collision or anyone who was travelling on the Belfast Road at the time of the collision to contact them in Enniskillen.
