Around 11,000 visitors are expected to descend on Belfast's Waterfront Hall for the chance to experience what the land of Westeros has to offer.
This year's display will focus on the storyline and events of series three of Game of Thrones with a few surprises from the fourth season.
It will feature over 100 original artefacts, including costumes, weapons, armour, set miniatures, storyboards and, of course, model dragons.
Fans will also get the chance to explore Kings Landing, sit on the coveted Iron Throne and 'climb the wall' at the new Oculus Rift interactive area.
HBO's Elana Loewenthal told UTV: "Everything in the exhibition is authentic, all of the costumes are made with real materials like furs, leathers, metals, so this is a truly immersive experience and its very different from the exhibit we had last year.
"About 80% of the items are different."
The hugely successful fantasy series is partly filmed in Northern Ireland, with various locations, including the Cushendun Caves, Murlough Bay, Ballintoy Harbour, and Larrybane, featuring on screen.
Northern Ireland has experienced a huge boost in tourism since the popular series aired on TV.
Martin Graham, from the Northern Ireland Tourist Board, said: "That big brand helps to get that Northern Ireland international stand out there where we would have difficulty in trying to achieve that through normal means.
"Those positive images gives it that real feel good factor so that people feel Northern Ireland looks beautiful with the castles, the countryside, and so on, from the viewers' perspective, but then they want to return and see that first hand as part of their holiday."
Game of Thrones is based on George R.R. Martin's best-selling books and is one of several high profile programmes and films that have been shot in Northern Ireland in recent years.
The Game of Thrones Exhibition will visit numerous locations around the world, including New York, Mexico, Austin, Rio de Janeiro, Oslo, Toronto, Vancouver and Belfast.
To mark the opening of the exhibition a number of Game of Thrones cast members attended a red carpet event at The Waterfront Hall on Tuesday night.
