It comes after publisher EA Sports announced the end of its 15-year association with Woods.
Now the bookmakers believe McIlroy, who has previously appeared on the game's cover, is poised to become the new face of the series.
He has been named as the 1/10 favourite to take over from Woods - despite a rough year which has seen him fail to win any competitions to add to his five titles in 2012.
Paddy Power said: "It's game over for Tiger as EA announced he will no longer be the headline act in their popular golf series.
"Despite Rory having a bad year on the course brand McIlroy is on the up and we think he is nailed on to elbow Woods off the cover of their next title."
In recent years the cover of the game has featured multiple golfing stars, with Woods sharing it alongside McIlroy and Rickie Fowler, and legends such as Arnold Palmer.
While the Ulsterman is out in front, bookies are also giving odds of 1/2 for an appearance by Fowler, 7/4 for Palmer and 5/2 for Portrush star Graeme McDowell.
A spokesperson for EA Sports said: "We do not comment on speculation and have nothing to announce at this time."
