Campbell claimed the title with a 14-11 victory in front of a 10,000 sell-out crowd at the ExCel arena.
The Team GB boxer landed a superb left-hand counter in the final round to floor his Irish opponent, who had beaten him in 2009 before the Hull man gained revenge with a close win in last year's World Championship semi-finals.
Nevin's tactics seemed to play into the hands of Campbell, who took a deserved 5-3 first-round lead.
An improved showing reduced the deficit to one point going into the final round, but Campbell made sure of the gold with a fine late effort, including the eye-catching counter that sent the Mullingar boxer to the canvas.
Team Ireland finish the Olympic Games with five medals in total - four in boxing, with Katie Taylor clinching gold in the women's lightweight division.
