McKillop, running for Ireland, romped home almost five seconds ahead of France's Lois Radius and Britain's Dean Miller.
The 24-year-old Glengormley runner's medal haul is Ireland's fourth of the games following Jason Smyth's sprint double gold earlier in the week.
Dame Tanni-Grey Thompson presented McKillop with flowers on the podium before being given his medal by proud mum Catherine.
It was a family celebration for the McKillop family as dad Paddy is Michael's coach.
The former St Malachy athlete has cemented his status as one of the world's finest Paralympians with five world and three Paralympic titles to his credit.
