In the game at Mourneview Park the defending champions found themselves a goal behind after just five minutes, when David Magee fired a low shot past Alan Blayney.
Parity was restored on 19 minutes when Glenavon failed to clear a Daryl Fordyce shot, allowing the ball to slide into the path of Jamie Mulgrew, who made no mistake from close range.
Nathan Hanley scored the winner in the 51st minute with a low shot past Andy Coleman.
Both sides finished the game with ten men when Robbie Garrett and Nathan Hanley were both shown straight red cards midway through the first half.
In the game at Ballyskeagh Alan Teggart capitalised on a defensive error to put Ballymena a goal up inside the opening 10 minutes, before GaryLiggett equalized for Distillery.
However, further goals from Gary McCutcheon (2) and Jordan Baker ensured an impressive opening day win for Roy Walker's side.
Elsewhere there was late drama at Seaview as Glentoran secured a 2-1 win over Crusaders.
Colin Coates' powerful header in the 79th minute cancelled out Colin Nixon's opener, but Darren Boyce struck in the 90th minute with a close-range header to seal the three points.
In Saturday's other games Carrick Rangers held out for 83 minutes against Cliftonville in their first game back in the top flight, but Martin Donnelly's close-range finish consigned the visitors to a 1-0 defeat.
Darren Murray scored on his debut for Donegal Celtic as they secured a 1-0 victory over Portadown, while the game between Dungannon Swifts and Coleraine ended goalless.
