Both teams are keen to progress to the semi-finals.
Alan McDonald's side overcame Institute on Saturday to move to top of the league table, one point ahead of Linfield who hold a game in hand.
Linfield will want to forget their disappointing defeat by Cliftonville at the weekend.
David Jeffrey has been issued with a three match touchline ban after being sent off in a recent league game with Dungannon Swifts, and will sit out the first of his three game suspensions on Tuesday night.
"Tuesday's certainly going to be a difficult game. The first leg will be forgotten about", Linfield's Kris Lindsay told the club's website.
"It's another "Big Two" game. It'll be a tough battle with the Glens coming off the back of a good victory up at Institute and us looking to banish the ghost of Saturday's result against Cliftonville."
"The Co-operative Insurance cup is a prestigious trophy and it's one we're very keen to be successful in," Linfield player Conor Hagan told the club's website.
"We've put ourselves in a good position by winning the first leg away. A 1-0 win at the Oval was a great result but I've no doubt the Glens will be coming out fighting on Tuesday night."
Meanwhile, Coleraine have a 3-2 advantage over Cliftonville from the away leg.
Portadown and Dungannon Swifts look like they have sealed their semi-final spots after the first legs.
The Ports lead 5-0 ahead of Ballinamallard's visit and the Swifts travel to Institute with the comfort of a 6-2 advantage.
