He had 41 seconds to spare over Cameron Donald at the chequered flag.
Race favourite John McGuinness, winner of 19 TT races, came third, overcoming a one-minute pit-lane penalty to set a new outright lap record on the final lap at 131.671mph.
Dunlop also posted a new personal best lap of the Mountain Course before sealing his fourth TT win.
He completed the six lap race at an average speed of 128.7mph.
It is 30 years since the late Joey Dunlop won on the famous circuit for the first time for Honda.
On Monday the Supersport race will be held in the morning, followed by the Superstock race in the afternoon.
