The men are all believed to have taken yellow speckled "Rolex" ecstasy tablets.
The party took place at a flat in the Thornhill Court area of Twinbrook on Friday evening.
A 41-year-old, named locally as Gerard Mulholland, was pronounced dead at the scene by emergency services. He was a successful DJ in the area.
Four men were taken to the Royal Victoria Hospital in Belfast for treatment after falling ill. Two of them remain in a stable condition, while two others have been released from hospital.
Detectives are investigating reports that yellow ecstasy type tablets, known as "speckled Rolex", may have been taken by the men during the house party.
Officers have urged anyone in possession of the pills not to take them, saying the pills should be disposed of "immediately and safely".
