Brothers Paul and Jeremy Eakin have seen their wealth mushroom to £310m due to wound care products from their company Eakin Holdings.
The wealthiest family in Northern Ireland, according to the paper, is that of Lord Ballyedmond, who was killed alongside three others in a helicopter crash in March.
His family's business - Norbrook Laboratories in Newry - made £540m last year, putting their wealth at £1.4bn.
This year's list revealed that Britain's richest people are wealthier than ever before with a combined fortune of £518.975bn, with a minimum of £85m needed to even be considered for the rankings.
The Queen added £10m to her personal fortune and is now ranked 285 with £330m, while TV chef Jamie and wife Jools Oliver saw their worth go up by £90m to £240m, ranking them at 396.
Philip Beresford, who compiles the list, said: "I've never seen such a phenomenal rise in personal wealth as the growth in the fortunes of Britain's 1,000 richest people over the past year.
"The richest people in Britain have had an astonishing year.
"While some may criticise them, many of these people are at the heart of the economy and their success brings more jobs and more wealth for the country."
