Part of the A4 Belfast Road, between Fivemiletown and Brookeborough, was closed after the incident on Friday evening.
The ambulance service has said that a total of nine people, including three children, have been taken to hospital with back and neck injuries.
Two people are believed to be in a serious condition, although their injuries are not thought to be life-threatening,
John McPoland, from the ambulance service, said 11 emergency crews were dispatched to the scene.
He said: "At the scene we found a number of patients who needed hospital treatment, and have taken them to the South West Acute Hospital and also Craigavon Area Hospital.
"I believe two of those patients are in a serious condition, although I don't think anyone has life-threatening injuries.
He added: "Because of the remote rural location, a number of local people in the area came out to the scene and attended to the casualties while the ambulance crews arrived and I would like to thank those people for their help."
Emergency services later stood down the major incident in Fivemiletown.
Diversions remained in place at Tempo on Friday night.
