The victim was walking in the area of Aspen Walk, Twinbrook, during the early hours of Friday when he was forced down an alleyway by his attacker.
Detectives are appealing for anyone who was in the area of Aspen Walk between 11pm on Thursday and 2am on Friday to contact the Rape Crime Unit on 0845 600 8000.
Cllr Brian Heading, SDLP Mayor of Lisburn, says the community is shocked and appalled.
"This depraved attack has stunned people in the Twinbrook area. It must have been such a horrific ordeal for the victim and our thoughts are with him at this difficult time," he said.
"Regrettably, this incident brings home the fact that male rape is an evil which exists within our communities.
He added: "Victims can carry the physical and mental scars of rape with them their whole lives and it is essential that better support services are put in place to help them."
