Army bomb experts were tasked to the scene at Finaghy Halt after a suspicious object was found on the railway line on Thursday.
Translink says the line between Great Victoria Street and Lisburn will "remain closed until further notice".
"Bus substitutions are operating for services to Lisburn and beyond," a spokeswoman said.
"Passengers should expect delays across the rail network."
Police said a number of homes have been evacuated in the areas of Finaghy Road North, Finaghy Park Central, Finaghy Park North, Ardmore Avenue and Diamond Gardens.
Finaghy Road North has been closed to traffic at its junction with Finaghy Park Central and Diamond Gardens.
Road closures are also in place at Finaghy Road North at its junction with Ardmore Park South.
The Lowe Memorial Church on the Upper Lisburn Road has been made available for people affected by the disruption.
A Red Cross bus has also been drafted in to provide additional assistance to residents.
