Jill Meagher, from Drogheda, was last seen leaving a bar in the city in the early hours of Saturday morning [Australian time].
She had been socialising with work colleagues, but failed to make it home.
Police are worried she may have "met with foul play."
Her handbag was found by officers close to the home she shares with her husband Thomas.
He raised the alarm after going out looking for her in the early hours of the morning.
Mrs Meagher had been living in Australia for three years, working for ABC Radio.
The disappearance is being investigated by the homicide department of Melbourne police.
They are carrying out searches and examining CCTV footage.
Detective Inspector John Potter, of Victoria Police, says they're concerned.
"The very circumstances of the case is worrying so as a result of that today the Homicide Squad Missing Persons Unit have taken over the case.
"We're hopeful for the best obviously, but we have some concerns that she may have met with foul play."
Her worried husband Thomas said:"I rang her mobile pretty much from 2 - 5 in the morning and there was no answer for a while and now the battery is flat."
Her uncle Michael McKeown added that his niece declined a lift home because she lived nearby and "knew the area well."
"She left that person who she worked with and hasn't been since."
