The new positions will be right across the store, including in the fresh food counters and customer services as well as the online department.
It will bring the total number employed at the Strand Road store to 300.
Sainsbury's will also recruit more than 150 seasonal roles across its stores in Northern Ireland to help cope with demand over the festive period. The major grocer hopes that many of these jobs will become permanent.
The upgrade work on the Strand Road store is due to be completed mid-November and will result in a one week closure next week, from 5 - 12 November. The revamped store will offer extended ranges in some areas including ladies clothing, cookware, and also a new pizza counter and redesigned bakery.
Nigel Macaulay, Regional Operations Manager for Sainsbury's, said: "We are delighted to be bringing additional employment to the north-west through our Strand Road store and indeed across Northern Ireland through our seasonal recruitment process.
"We have been listening to the needs of our customers in the northwest and it is fantastic to be able to now provide the Derry/Londonderry area with an online shopping service which will be launching on 20th November as well as an extended and upgraded store offering."
