Dornan, 31, was named as the frontrunner for the part after actor Charlie Hunnam recently bowed out of the project earlier this month over his television commitments and filming schedule.
Other actors in the running for the highly coveted role of billionaire Christian Grey included Ryan Gosling, Alexander Skarsgård, Ian Somerhalder and Matt Bomer.
According to reports Dornan was given the part after screen testing with female lead Dakota Johnson, who will play timid student Anastasia Steele.
Shooting on the film is expected to begin on 1 November.
Dornan, who previously dated actress Keira Knightley, has recently thrilled audiences in dark crime drama The Fall as serial killer Paul Spector.
He has also starred as Sheriff Graham Humbert in the popular US series Once Upon a Time.
Dubbed "mummy porn", Fifty Shades by author EL James went on to become the fastest-selling paperback of all time.
The film, directed by Sam Taylor-Johnson, is due out in August next year.
Dornan and his singer-songwriter wife Amelia Warner, who he married in April, are currently expecting their first child.
Reaction to the casting announcement has been largely positive with many fans commenting that the actor is perfect for the role.
Jamie Dornan was trending worldwide on microblogging site Twitter within hours of the news breaking, and many users offered their congratulations to the Holywood man.
Dornan's onscreen wife in The Fall, Bronagh Waugh wrote: "I think my onscreen husband @JamieDornan1 has just landed THE biggest role of the CENTURY!!?! How will I ever be naked with him again?!!!"
Co-star Gerard McCarthy said: "Absolutely ecstatic for @JamieDornan1! Not only a fine actor, but moreover, a true gent! Well deserved matey!"
So far the actor himself has remained quiet about his latest casting news.
