The famous Glaswegian, known to many as The Big Yin, is expected to receive the Freedom of the City following a vote by councillors next month.
Lord Provost Bob Winter is calling for the city to recognise the performer for his contribution to comedy, film, music and charities.
Connolly was born and raised in Glasgow where he began his working life in the shipyards, which he said was great training for life as a comedian.
Now a veteran stand-up after more than 30 years on the stage, Connolly has appeared in numerous films and TV series.
Mr Winter said: "Billy Connolly is arguably the world's best-known Glaswegian and is truly deserving of the Freedom of the City.
"A true Scottish and Glasgow great, as well as a Hollywood film star, he also works selflessly for charity, including Comic Relief.
"More recently, he has confronted difficult issues stemming from his personal background, such as childhood abuse and later alcoholism, with unflinching honesty.
"But above all, Billy Connolly has presented himself as a proud citizen of the city of Glasgow. In doing so he has shown the world the unique humour, generosity and resilience of Glaswegians."
The Lord Provost also praised Connolly's contribution to local causes.
He staged benefit concerts to help uninsured residents during the east end floods in 2002, and raised money for a hospice with a charity cycle ride from Glasgow to London, Mr Winter said.
A motion will be submitted to the city council at a meeting on June 24.
Past recipients of the Freedom of the City include Nelson Mandela and Sir Alex Ferguson.
A spokeswoman for Connolly did not wish to comment.
